Victoria Police Superintendent David Clayton said when police arrived at the scene they were confronted by a man brandishing a knife and threatening them.

A police spokesman later said that authorities are not looking for any other offender, adding that the investigation is at the "very early stage", and that there is "no known link to terrorism" so far.

"Three people have been stabbed".

'The exact circumstances are yet to be determined at this stage.

The centre of Melbourne's CBD is in lock down.

Images from the scene show a large white sheet covering a section of the road near what appears to be blood. Only the soles of the person's feet were visible.

Members of the public are being urged to avoid the area with several streets reportedly placed on lock-down.

Trams along the Bourke and Swanston Street routes are being either held up or diverted around the affected area.

Terror sirens have been activated from loudspeakers in the city.

Police said the scene had been cordoned off, and it was being "made safe" by a bomb response unit.

A second person suffered a head injury and has been taken to hospital.

It's understood a third person has also been taken to hospital.

The officers then retreated to the other side of the road as the attacker, dressed in a long jacket and grey trousers, ominously pursued them as horrified bystanders called on police to shoot him.

Police are asking people to avoid the area and said more information would be given when it was available. They said they heard screams and cries from the crowd before they were rushed from the area.

One witness, Sarah Krug said she was heading to the night noddle markets when "all hell broke loose".

SBS News has contacted Victoria Police for comment. Some witnesses said the auto exploded before the man began stabbing people.

"I didn't know what was happening there was just so much chaos and screaming".

Chilling footage uploaded to social media shows a man attacking police with a knife moments after the vehicle exploded.

Another witness said: "Big fire".
