The Falcon Heavy rocket exerts 5.1 million pounds of thrust - that of more than a dozen jetliners, SpaceX said.
The rocket was launching a satellite into orbit for Saudi Arabian company Arabsat.
The historic launch was watched by millions around the globe thanks to Mr Musk's body decision to mount a cherry-red Tesla sports vehicle as the rocket's payload.
The Falcon-Heavy rocket was launched at 10.35 pm.
SpaceX aims to land two of the first-stage boosters back at Cape Canaveral, just like it did for the rocket's debut a year ago.
The Arabsat 6A is a modern communications satellite developed by Lockheed Martin for Arabsat, an organisation founded by the Arab League in 1976 to provide telecommunications services to the region. SpaceX then recovered all three of the boosters back on Earth, achieving a feat it wasn't able to pull off in the rocket's demonstration flight a year ago. During the heavy-lift rocket's debut launch previous year, the first attempt to recover the center core failed.
On Wednesday Elon Musk, SpaceX CEO suggested that a delay was likely due to "upper atmospheric wind shear" and SpaceX confirmed there would be no launch.
SpaceX typically launches Falcon 9 rockets. This is all part of SpaceX's plan to refurbish rocket boosters so they can be used again, lowering the cost of spaceflights.
